The purpose of an elutriation rotor is to size-separate and/or size-concentrate monodisperse suspensions of single cells or particles . Each cell in the chamber is affected by the two opposing forces as a result of the combination of two separation technologies in an elutriation rotor:It is being pushed away from the axis of rotation by centrifugal force.
This is sedimentation occurring as a result of a centrifugal force field.Driving the fluid in the direction of the axis of rotation—this is known as “counterflow elutriation” This is the washing-based separation method.In an elutriation chamber with a funnel form, separation occurs.
A suspension of cells is injected into the centrifuge’s rotor at a predetermined flow rate from outside the machine while it is spinning.
Small cells with slow rates of sedimentation rapidly wash in the direction of rotation. These cells are flushed from the chamber into a collection jar after passing up through the rotor. Cells that are a little bit bigger or denser go through the chamber more slowly and come to rest at the elutriation boundary.
The largest or densest cells stay close to the chamber’s inlet where there is a high centrifugal force and fluid velocity. Successive fractions of increasingly large or dense cells can be washed out of the rotor and collected by gradually raising the flow rate. All cells will eventually be eluted from the chamber with a steady increase in flow rate.
